Overview

**CVM-1118 + nivolumab** is an investigational combination therapy for advanced cancers, currently in clinical trials for unresectable hepatocellular carcinoma (HCC). CVM-1118 (foslinanib) is a novel small molecule targeting tumor cell survival and vasculogenic mimicry (VM) by binding to TNF receptor associated protein 1 (TRAP1, also known as Hsp75), inducing mitochondrial apoptosis, arresting cell cycle at G2/M phase, and inhibiting VM network formation[1][2][4]. CVM-1118 is rapidly metabolized to its active metabolite CVM-1125. Nivolumab is a human monoclonal IgG4 kappa antibody that acts as a checkpoint inhibitor targeting PD-1, enhancing T-cell anti-tumor responses. The combination aims to leverage the complementary mechanisms of apoptosis induction and immune checkpoint inhibition for improved anti-tumor efficacy. Developed and tested by TaiRx, CVM-1118 is supported by favorable early safety and efficacy data in advanced HCC compared to other regimens such as atezolizumab plus bevacizumab[1][3].
