Overview

CVN058 is a novel, brain-penetrant small molecule that acts as a potent and selective antagonist of the serotonin type 3 receptor (5-HT3R). It is being developed by Cerevance for the treatment of cognitive impairment associated with schizophrenia (CIAS). The drug has demonstrated efficacy in rodent models of cognition and was well-tolerated in Phase 1 studies. In a Phase 1b clinical trial involving patients with schizophrenia or schizoaffective disorder, a single oral dose of CVN058 led to statistically significant improvements in mismatch negativity (MMN), an auditory evoked potential biomarker generated in the cerebral cortex that is typically impaired in schizophrenic patients. This suggests that CVN058 may help correct excitatory/inhibitory imbalance and improve aspects of cognition in these individuals[1][3][6].
