CVX-045 is a recombinant humanized monoclonal antibody fused to two thrombospondin 1 (TSP-1) mimetic nonamer peptides via a proprietary linker. It acts as an antiangiogenic and antineoplastic agent by mimicking the activity of TSP-1, an endogenous inhibitor of angiogenesis. The drug inhibits endothelial cell migration and angiogenesis in vitro and in vivo, showing significant anti-tumor activity in preclinical models both alone and in combination with chemotherapy or targeted therapies. Developed originally by CovX and later by Pfizer, CVX-045 was investigated for the treatment of advanced solid tumors but development was discontinued after phase 1 clinical trials due to strategic reasons rather than safety concerns[1][2][3][4][5][6].
