Overview

CWP232291 is a small molecule peptidomimetic and prodrug developed by JW Pharmaceutical and the Korea Research Institute of Bioscience and Biotechnology for oncology indications. It acts as an inhibitor of the Wnt/β-catenin signaling pathway, leading to degradation of β-catenin, downregulation of target genes such as cyclin D1 and survivin, and induction of apoptosis in cancer cells. Upon administration, it is rapidly converted in serum to its active form, CWP232204, which binds to Sam68 (KHDRBS1), modulating alternative splicing events that favor pro-apoptotic isoforms. The drug also inhibits molecular chaperone GRP78 protein. Preclinical studies have shown efficacy against hematologic malignancies (including acute myeloid leukemia) and solid tumors such as gastric cancer; it has demonstrated activity even in chemotherapy-resistant models[1][4][5][6][7].
