CX-2005

CX-2005 is a preclinical-stage Probody drug conjugate (PDC) developed by CytomX Therapeutics that targets CD71 (transferrin receptor protein 1). CD71 is highly and homogeneously expressed across multiple solid tumors and lymphomas, making it an attractive target for intracellular payload delivery. However, its expression on normal tissues, particularly hematopoietic progenitor cells, has historically limited the development of traditional antibody-drug conjugates (ADCs) due to severe toxicities. CX-2005 utilizes CytomX's Probody technology, featuring a peptide mask linked to the anti-CD71 antibody via a protease-cleavable substrate. This mask remains intact in circulation and normal tissues, preventing binding to CD71, but is proteolytically cleaved by tumor-associated proteases in the tumor microenvironment to selectively release the active antibody. In preclinical studies, CX-2005 demonstrated complete tumor regressions in mouse models of lymphoma, breast cancer, and lung cancer, while avoiding the life-threatening hematological toxicities observed with unmasked anti-CD71 ADCs in non-human primates.
