CX-2029 is a Probody drug conjugate (PDC), also described as an antibody-drug conjugate (ADC), designed to target CD71, the transferrin receptor 1. It consists of a protease-cleavable, masked humanized monoclonal antibody that binds to CD71 only after being unmasked by tumor-associated proteases in the tumor microenvironment. The antibody is conjugated via a valine–citrulline linker to monomethyl auristatin E (MMAE), a potent microtubule inhibitor. This design allows for selective delivery of MMAE to tumors while sparing normal tissues, addressing previous safety concerns with targeting CD71 directly. CX-2029 has demonstrated antitumor activity in preclinical models and has shown promising clinical activity at tolerable doses in patients with advanced malignancies, particularly those with squamous histologies[1][2][3][4][5][6].
