CX-2043 is a conditionally activated **antibody-drug conjugate (ADC)** utilizing Probody technology to target **EpCAM (epithelial cell adhesion molecule)**, which is highly expressed in epithelial cancers including colorectal, ovarian, uterine, stomach, and lung cancers. The drug consists of an EpCAM-targeting masked antibody conjugated via lysine residues and a protease-cleavable peptide linker to a novel maytansinoid (DM21L) cytotoxic payload, designed for enhanced bystander effect and stability. The Probody approach restricts activation to the tumor microenvironment, minimizing healthy tissue toxicity compared to conventional EpCAM ADCs. In preclinical studies, CX-2043 showed potent cytotoxicity, efficacy in xenograft models, and a favorable safety profile in comparison to unmasked EpCAM ADCs. The mechanistic action involves inhibition of tubulin polymerization, thereby arresting mitosis and inducing tumor cell death[1][2][3][10][11].
