Overview

CX-659S is a novel diaminouracil derivative with potent antioxidative and anti-inflammatory properties. It acts as a lipid peroxidation inhibitor and demonstrates strong scavenging activity against hydroxyl radicals and peroxynitrite. CX-659S inhibits lipid peroxidation in vitro and reduces acute skin inflammation in animal models by attenuating the accumulation of lipid peroxides. Its mechanism involves indirect inhibition of Langerhans cell function through blockade of the MEK1/2–ERK1/2 pathway in keratinocytes, leading to reduced expression of co-stimulatory molecules (CD80/CD86) on Langerhans cells and decreased production of proinflammatory cytokines such as IL‑4 and IL‑10. Unlike corticosteroids or cyclosporine A, CX‑659S does not cause immunosuppression or typical steroid-associated side effects. It has shown efficacy in preclinical models for contact hypersensitivity reactions (CHRs) and atopic dermatitis-like conditions[1][4][5][6]. Developed by Japan Energy.
