CX-717 is a small molecule ampakine compound developed by Cortex Pharmaceuticals (now RespireRx Pharmaceuticals) and first synthesized in 1996. It acts as a positive allosteric modulator of AMPA-type glutamate receptors, enhancing synaptic transmission at cortical synapses that use glutamate as a neurotransmitter. This mechanism is believed to promote synaptic plasticity and improve cognitive performance, memory formation, and learning. Preclinical and clinical studies have investigated its potential for treating attention-deficit/hyperactivity disorder (ADHD), Alzheimer's disease, respiratory insufficiency (notably opioid-induced respiratory depression), and sleep disorders. Although oral formulations were limited by poor bioavailability and blood-brain barrier penetration, intravenous forms continue to be explored for respiratory indications[1][2][3][4].
