CX1106 is a small molecule inhibitor of thymidylate synthase (TS), an enzyme critical for the synthesis of thymidine nucleotides required for DNA replication and cell division. By inhibiting TS, CX1106 reduces thymine nucleotide synthesis, leading to inhibition of DNA synthesis and cell division, causing DNA damage and tumor cell apoptosis. Unlike other TS inhibitors such as pemetrexed or fluorouracil, CX1106 does not require active transport into cells nor polyglutamation for its antitumor activity. It was developed as a potential antitumor agent primarily targeting neoplasms such as advanced head and neck squamous cell carcinoma[2][3][4].
