CX1763 is a preclinical, low-impact ampakine small molecule that acts as a positive allosteric modulator of AMPA-type ionotropic glutamate receptors, enhancing glutamate-evoked synaptic currents and hippocampal excitatory postsynaptic potentials in vivo without inducing epileptiform activity at therapeutically relevant exposures.[1][3][7][10] Developed originally by Cortex Pharmaceuticals and later included in a Biovail/RespireRx ampakine portfolio, CX1763 has shown in rodent models to increase measures of attention in the five-choice serial reaction time task, reduce amphetamine-induced hyperactivity relevant to ADHD, and contribute (via its water-soluble prodrug CX1942) to reversal of opioid-induced respiratory depression, supporting potential applications in neuropsychiatric and respiratory indications.[1][3][6][9][10][12]
