Overview

**CX1942** is a water-soluble glycine ester prodrug of **CX1763**, a low-impact ampakine that allosterically potentiates AMPA glutamate receptors (AMPARs) to enhance glutamate-elicited currents without blocking receptor desensitization, minimizing risks like epileptogenicity associated with high-impact ampakines. Developed by **RespireRx Pharmaceuticals**, it rapidly hydrolyzes in blood (half-life 2-15 min across species) to release the active CX1763, which boosts hippocampal synaptic transmission, improves attention in ADHD models, reduces amphetamine-induced hyperactivity, and potently reverses opioid-induced respiratory depression (OIRD) at 2.5-10 mg/kg IV in rats. CX1942 offers advantages in solubility for IV formulations and shows a wide safety margin, with CX1763 lacking epileptogenic effects up to 1500 mg/kg orally in rats.[1]
