CX390 is a bispecific chimeric antigen receptor (CAR)-T cell therapy designed to simultaneously target B-cell maturation antigen (BCMA) and CD19. Developed by Caring Cross and Vector BioMed, the construct utilizes a bicistronic format featuring a bi-paratopic humanized VHH-based BCMA binder and a human single-chain variable fragment (scFv) targeting CD19, driven by an EF1a promoter. This duoCAR approach aims to provide comprehensive depletion of the B-cell lineage, from early B-cell precursors to terminal plasma cells, to prevent relapse in multiple myeloma and treat B-cell-mediated autoimmune diseases. Preclinical studies have demonstrated superior CAR expression, potent cytotoxicity, and sustained anti-tumor persistence.
