Drug intelligence / Profile preview
Cx43-M1 (also known as Cx43(M1) or MHC1) is a first-in-class monoclonal antibody that selectively blocks connexin 43 (Cx43) hemichannels without disrupting gap junction communication. Developed by the laboratory of Dr. Jean X. Jiang at the University of Texas Health Science Center at San Antonio (UT Health San Antonio), the antibody targets the second extracellular loop of Cx43. By inhibiting the pathological opening of Cx43 hemichannels, Cx43-M1 prevents the release of pro-inflammatory and neurotoxic molecules such as ATP and glutamate. The antibody has demonstrated therapeutic potential in preclinical models of osteoarthritis (OA), acute spinal cord injury (SCI), optic nerve injury, and bone loss. AlaMab Therapeutics licensed the technology to develop a humanized version, ALMB-0166, which is currently in clinical development for acute spinal cord injury.
