CXC195 is a novel small molecule derivative of tetramethylpyrazine (TMP) currently under preclinical investigation for its neuroprotective properties, particularly in the treatment of Parkinson's disease. It functions primarily by inhibiting apoptosis and modulating the PI3K/Akt/GSK3β signaling pathway. In animal models of Parkinson's disease, CXC195 has demonstrated the ability to protect dopaminergic neurons from 6-hydroxydopamine (6-OHDA)-induced damage. Its mechanism involves decreasing the expression of pro-apoptotic proteins such as Bax and cleaved caspase-3, while increasing the levels of the anti-apoptotic protein Bcl-2. Furthermore, CXC195 restores the phosphorylation of Akt and inhibits the activity of GSK-3β (by increasing its phosphorylation at Ser9), which are critical steps in promoting neuronal survival and mitigating neurodegeneration.
