Overview

CXCL12-modified oncolytic adenovirus is an engineered, replication-competent oncolytic adenovirus serotype 5 (Ad5) designed for cancer therapy, particularly breast cancer. Developed by researchers at Louisiana State University, the virus is genetically modified to incorporate the human chemokine ligand CXCL12 (SDF-1) fused to a T4 fibritin trimerization domain within the tail domain of the adenovirus fiber protein. This modification retargets the virus to selectively infect and lyse cancer cells overexpressing the chemokine receptors CXCR4 and CXCR7, bypassing the need for the native coxsackievirus and adenovirus receptor (CAR/CXADR) which is often downregulated in advanced tumors. Upon infection, the virus replicates within the cancer cells, leading to oncolysis and the release of progeny virions to propagate the therapeutic effect throughout the tumor stroma.
