CXCL122 is an engineered, dimerized variant of the chemokine CXCL12 (also known as stromal cell-derived factor 1 or SDF-1). It is designed to function as a biased agonist at the C-X-C chemokine receptor type 4 (CXCR4), a target that is frequently overexpressed in head and neck squamous cell carcinoma (HNSCC) and linked to tumor progression and treatment resistance. Unlike the native monomeric form of CXCL12, which often promotes tumor proliferation and metastasis, CXCL122 has been shown to inhibit HNSCC cell proliferation and enhance the therapeutic efficacy of radiotherapy. This anti-tumor activity is associated with the upregulation of the KISS1R (kisspeptin receptor) pathway. Preclinical studies indicate that CXCL122 effectively localizes to tumor sites and works synergistically with radiation to suppress tumor growth in xenograft models.
