Overview

CXCL13-EcN is an engineered live biotherapeutic product (LBP) based on the probiotic bacterium *Escherichia coli* Nissle 1917 (EcN). It has been synthetically modified to express the human chemokine CXCL13, a key regulator of B cell recruitment and germinal center (GC) formation. In the context of bladder cancer, CXCL13-EcN is designed for intravesical administration to colonize the tumor microenvironment and stimulate the development of GCs in tumor-draining lymph nodes. This localized expression of CXCL13 aims to amplify tumor-specific antibody responses and enhance the efficacy of immune checkpoint inhibitors, such as PD-1 blockades. Preclinical studies indicate that this approach can lead to long-term survival and protective immunity against tumor rechallenge by remodeling the intratumoral microbiome and boosting adaptive humoral immunity.
