CXCL14, also known as BRAK (Breast and Kidney-expressed chemokine), is a homeostatic chemokine that functions as a potent tumor suppressor and immune modulator. It is constitutively expressed in normal epithelial tissues but is frequently downregulated or lost during the progression of various malignancies, including head and neck squamous cell carcinoma (HNSCC), lung cancer, and melanoma. CXCL14 acts by recruiting and activating natural killer (NK) cells, dendritic cells, and monocytes to the tumor microenvironment, thereby enhancing anti-tumor immunity. It also exhibits anti-angiogenic properties. Research indicates that restoring CXCL14 expression through gene therapy (forced expression) or the administration of recombinant protein can significantly inhibit tumor growth and suppress metastasis. The anti-metastatic effect of CXCL14 is primarily mediated by its ability to mobilize NK cells, which are indispensable for its therapeutic efficacy in preclinical models.
