Overview

CXCR4 modified anti-BCMA CAR T cells are a form of autologous chimeric antigen receptor (CAR) T cell therapy engineered to express both an anti-B-cell maturation antigen (BCMA) CAR and the chemokine receptor CXCR4. The addition of the BCMA-targeted CAR enables these T cells to specifically recognize and kill malignant plasma cells expressing BCMA, a marker highly expressed in multiple myeloma. Co-expression of the chemokine receptor CXCR4 enhances trafficking and homing of these engineered T cells to the bone marrow microenvironment, where multiple myeloma primarily resides. This dual modification aims to improve both tumor targeting and infiltration into disease sites, potentially increasing therapeutic efficacy for patients with relapsed or refractory multiple myeloma[2][3][7]. Early-phase clinical trials are evaluating their safety and tolerability in this patient population[4].
