Overview

CXCR4-modified CD19 CAR-T cells are an investigational autologous cell therapy being developed for the treatment of relapsed or refractory B-cell malignancies. The therapy involves T cells that have been genetically engineered to express a chimeric antigen receptor (CAR) specific for the CD19 antigen, which is widely expressed on B-cell leukemia and lymphoma cells. Additionally, these CAR-T cells are modified to overexpress the chemokine receptor CXCR4. This modification is designed to exploit the CXCR4-CXCL12 signaling axis, thereby enhancing the trafficking of the CAR-T cells to tumor-rich environments and improving their homing to the bone marrow. Early clinical data from a Phase I trial (NCT04684472) led by Liqun Zou has demonstrated preliminary efficacy and safety in patients with B-cell acute lymphoblastic leukemia (B-ALL) and B-cell non-Hodgkin lymphoma (B-NHL).
