Drug intelligence / Profile preview
CXCR4R334X-engineered NK cells are an experimental, preclinical cell therapy consisting of ex vivo expanded human natural killer (NK) cells genetically engineered via mRNA transfection to transiently express the gain-of-function CXCR4 R334X variant (associated with WHIM syndrome). This modification is designed to enhance the homing capacity of the adoptively transferred NK cells to the bone marrow by targeting the CXCR4/SDF-1alpha (CXCL12) chemokine axis. Developed by researchers at the Karolinska Institutet and the National Institutes of Health (NIH), this therapy is being investigated for the treatment of bone-marrow-residing hematological malignancies, such as acute myeloid leukemia (AML) and multiple myeloma.
