CXCR5-EGFR CAR-T is a genetically engineered autologous T cell therapy that co-expresses an epidermal growth factor receptor (EGFR)-targeting chimeric antigen receptor (CAR) and the chemokine receptor CXCR5. The CAR targets EGFR, a tumor-associated antigen commonly overexpressed in non-small cell lung cancer (NSCLC) and certain other solid tumors. CXCR5 enables chemotactic migration of the CAR-T cells toward CXCL13, a chemokine upregulated in CXCL13-positive tumors, thereby improving infiltration into tumor sites and facilitating efficient tumor cell killing. This design addresses the limitation of poor CAR-T cell trafficking in solid tumors and seeks to reduce off-tumor toxicity by limiting T cell localization and activation to tumors that are both EGFR and CXCL13 positive. The therapy is currently being investigated primarily for NSCLC.
