Overview

CXCR5-EGFR CAR-T cells are autologous, genetically engineered T cells expressing a chimeric antigen receptor (CAR) targeting **epidermal growth factor receptor (EGFR)** and co-expressing **C-X-C chemokine receptor type 5 (CXCR5)**. The CXCR5 modification enables enhanced migration of CAR-T cells toward tumors that secrete its ligand CXCL13, which is overexpressed in non-small cell lung cancer (NSCLC). This design is intended to increase tumor infiltration, improve cytotoxic activity against EGFR-positive tumor cells, and potentially reduce off-tumor, on-target toxicity. The therapy is under evaluation for the treatment of advanced adult non-small cell lung cancer, specifically in EGFR-positive patients [1][3][5][7][9]. Primary mechanism is immunologic cytotoxicity mediated by engineered T lymphocyte replacement.
