CXCR5 modified EGFR CAR-T cells are a form of autologous chimeric antigen receptor (CAR) T cell therapy engineered to target tumors expressing the epidermal growth factor receptor (EGFR). These T cells are further genetically modified to express the chemokine receptor CXCR5, which enhances their migration toward tumor sites that secrete its ligand, CXCL13. This dual modification aims to improve infiltration and cytotoxicity against solid tumors—particularly non-small cell lung cancer (NSCLC)—that co-express both EGFR and high levels of CXCL13. The mechanism involves specific recognition and killing of EGFR-positive tumor cells by the CAR construct while the added expression of CXCR5 facilitates homing to the tumor microenvironment rich in CXCL13. Preclinical studies have shown improved efficacy in eradicating such tumors compared to conventional anti-EGFR CAR-T therapies[1][2][4]. Clinical trials are ongoing for advanced NSCLC[3][5][8].
