CXCR5-modified EGFR chimeric antigen receptor T cells are an investigational autologous cell therapy designed to enhance the efficacy of CAR-T cells against solid tumors by improving their trafficking and infiltration. These T cells are genetically engineered to express a chimeric antigen receptor (CAR) targeting the epidermal growth factor receptor (EGFR), a protein frequently overexpressed in various epithelial malignancies including non-small cell lung cancer (NSCLC) and glioblastoma. To overcome the physical and biochemical barriers of the tumor microenvironment, the cells are further modified to co-express the chemokine receptor CXCR5. This receptor allows the CAR-T cells to migrate toward higher concentrations of the chemokine CXCL13, which is often secreted within tumor tissues or tertiary lymphoid structures. By combining targeted cytotoxicity against EGFR-positive cells with improved site-specific homing, this therapy aims to increase the local density of effector cells at the tumor site and achieve better therapeutic outcomes than conventional CAR-T approaches.
