CXD101 + nivolumab

CXD101 + nivolumab is a combination therapy under investigation for the treatment of advanced or metastatic microsatellite-stable colorectal cancer (MSS CRC). CXD101 is an oral small molecule inhibitor of class I histone deacetylases (HDACs), which modulates gene expression and can enhance tumor immunogenicity. Nivolumab is a fully human IgG4 monoclonal antibody that targets programmed cell death protein 1 (PD-1), an immune checkpoint receptor on T-cells, thereby blocking its interaction with PD-L1 and PD-L2 to restore anti-tumor immune responses. The combination aims to increase the effectiveness of immunotherapy in MSS CRC, a population typically resistant to checkpoint inhibitors alone. In phase Ib/II clinical trials, this regimen demonstrated tolerability and signs of efficacy in heavily pretreated patients[1][2][3].
