Overview

CXR101 is a preclinical-stage gene therapy developed by CorriXR Therapeutics, designed to treat solid tumors including lung squamous cell carcinoma and head and neck squamous cell carcinoma. The therapy utilizes a proprietary CRISPR/Cas gene editing system encapsulated in lipid nanoparticles (LNPs) for localized delivery. CXR101 specifically targets and knocks out the NRF2 (Nuclear factor erythroid 2-related factor 2) transcription factor, which is a master regulator of the cellular antioxidant response. In many squamous cell carcinomas, NRF2 is overexpressed, driving a pro-oncogenic tumor microenvironment and conferring resistance to chemotherapy and radiotherapy. By disrupting NRF2 through direct intratumoral or local injection, CXR101 aims to reprogram the tumor microenvironment and sensitize tumor cells to standard-of-care oncological treatments.
