Overview

CY3018 is a differentiated, next-generation, central nervous system (CNS)-penetrant soluble guanylate cyclase (sGC) stimulator developed by Cyclerion Therapeutics. It is designed to preferentially localize to the brain and exhibits a pharmacology profile that suggests potential for the treatment of neuropsychiatric diseases and disorders. Preclinical data indicate that CY3018 achieves higher CNS exposure compared to earlier sGC stimulators such as CY6463, with an increased cerebrospinal fluid (CSF) to plasma ratio and greater pharmacological activity in the CNS relative to peripheral tissues. The drug acts by stimulating sGC in the brain, thereby enhancing nitric oxide (NO)-sGC-cyclic guanosine monophosphate (cGMP) signaling—a pathway implicated in regulating mitochondrial function, neuronal activity, inflammation, and vascular dynamics within the CNS. As of June 2025, CY3018 remains in preclinical development for central nervous system diseases[1][2][3][4][7][8][9].
