Cy5-uPA-probe is a fluorescently labeled activity-based probe designed for the non-invasive near-infrared (NIR) imaging of active urokinase plasminogen activator (uPA) expression in vivo. Developed by researchers at the University of Antwerp, the probe is based on the irreversible, non-peptidic uPA inhibitor UAMC-00150 conjugated to a Cy5 fluorophore. It selectively and covalently binds to active uPA, a serine protease heavily implicated in extracellular matrix degradation, tumor invasion, and metastasis. Cy5-uPA-probe has been evaluated in preclinical orthotopic breast cancer mouse models (such as MDA-MB-231) to visualize uPA activity, serving as a potential diagnostic biomarker tool for breast cancer and other solid tumors.
