Overview

CYAD-02 is an autologous, next-generation chimeric antigen receptor T cell (CAR-T) therapy developed for the treatment of relapsed or refractory acute myeloid leukemia (AML) and myelodysplastic syndromes (MDS). The therapy uses a patient's own T-cells that are genetically engineered to express a CAR based on the natural killer group 2D (NKG2D) receptor. This enables the modified T-cells to recognize and kill tumor cells expressing NKG2D ligands. Additionally, CYAD-02 incorporates shRNA technology to silence expression of the endogenous NKG2D ligands MICA and MICB on the engineered T-cells themselves, which enhances their expansion and persistence in vivo. The mechanism of action involves direct cytotoxicity against cancer cells as well as targeting immunosuppressive cells in the tumor microenvironment that express NKG2D ligands[1][6][7]. Development was led by Celyad Oncology.
