Overview
A multi-agent investigational combination therapy consisting of **CYAD-101** (an allogeneic, non-gene edited, NKG2D-based CAR-T cell therapy with TCR Inhibitory Molecule for reduced graft-versus-host disease), **leucovorin** (a folate analog used to enhance 5-FU activity), **fluorouracil** (a pyrimidine analog antimetabolite chemotherapy), **oxaliplatin** (a platinum-based chemotherapy agent), and **pembrolizumab** (a checkpoint inhibitor antibody targeting PD-1). This combination has been studied in clinical trials for the treatment of refractory metastatic colorectal cancer, leveraging conceivable immune synergy between allogeneic CAR-T cells, cytotoxic chemotherapy, and immune checkpoint inhibition. All drugs, except the CAR-T component, are standard-of-care agents in mCRC; the combination aims to overcome immune suppression in solid tumors through mechanistic complementation[1][3][5][7].
