**cyclic GMP-AMP** is a cyclic purine dinucleotide composed of AMP and GMP units cyclized via 3',5'-linkages[1]. It is an endogenous second messenger produced by the enzyme cyclic GMP-AMP synthase (cGAS) when cGAS detects abnormal cytoplasmic DNA[4][6]. Once generated, cyclic GMP-AMP binds to the stimulator of interferon genes (STING) protein, activating a signaling pathway that results in type I interferon and cytokine production, thereby triggering a robust innate immune response[4][6]. Cyclic GMP-AMP has notable roles in antiviral defense, cancer immunotherapy (as a STING agonist), and is under investigation as a therapeutic agent and vaccine adjuvant[6][8].
