Drug intelligence / Profile preview
Cyclic guanosine monophosphate–adenosine monophosphate (cyclic GMP-AMP or cGAMP) is an endogenous cyclic dinucleotide and second messenger that plays a pivotal role in the innate immune system. Synthesized by the cytosolic DNA sensor cyclic GMP-AMP synthase (cGAS) in response to the presence of double-stranded DNA in the cytoplasm, cGAMP binds to and activates the stimulator of interferon genes (STING) receptor. This activation triggers a downstream signaling cascade involving TANK-binding kinase 1 (TBK1) and interferon regulatory factor 3 (IRF3), leading to the production of type I interferons and other pro-inflammatory cytokines. cGAMP is widely utilized as a research reagent and is being investigated preclinically as a vaccine adjuvant and immunotherapeutic agent for infectious diseases and cancer.
