Overview

Cyclic RGD peptides are a class of synthetic cyclic peptides containing the arginine-glycine-aspartic acid (RGD) tripeptide motif. They are designed to mimic the cell adhesion domain of extracellular matrix proteins like fibronectin and vitronectin, allowing them to bind with high affinity and selectivity to integrins, particularly integrin alpha-V/beta-3 (αvβ3) and integrin alpha-V/beta-5 (αvβ5). These integrins are highly overexpressed on angiogenic endothelial cells and various tumor cells, making cyclic RGD peptides valuable tools for targeted drug delivery (such as in liposomes or nanoparticles) and as molecular imaging probes (when radiolabeled) for cancer detection and monitoring.
