Cyclin B1 peptide-pulsed autologous dendritic cell vaccine is an investigational immunotherapy designed to elicit a cytotoxic T-lymphocyte (CTL) response against tumor cells that overexpress cyclin B1. Cyclin B1 (CCNB1) is a regulatory protein essential for the G2/M phase transition of the cell cycle; while it is expressed at low levels in normal cells, it is significantly overexpressed in various malignancies, including non-small cell lung cancer (NSCLC), breast cancer, and colorectal cancer. The vaccine is produced by harvesting a patient's own peripheral blood mononuclear cells via leukapheresis, differentiating them into dendritic cells (DCs) in vitro, and pulsing them with MHC-restricted cyclin B1 peptides. These antigen-loaded dendritic cells are then re-administered to the patient, where they function as professional antigen-presenting cells to prime and expand cyclin B1-specific T-cells, potentially leading to the targeted destruction of cancer cells.
