Cyclin D1 siRNA (also referred to as CyD1 siRNA or siCyD1) is a research-stage RNA interference (RNAi)-based therapeutic modality designed to silence the expression of Cyclin D1 (CyD1), a key cell cycle-regulatory protein encoded by the CCND1 gene. By degrading CyD1 mRNA, this small interfering RNA reduces Cyclin D1 protein levels, leading to the hypophosphorylation of retinoblastoma protein (RB) and subsequent cell cycle arrest in the G1 phase. Because Cyclin D1 is frequently overexpressed in various malignancies and inflammatory conditions, cyclin D1 siRNA has been investigated for the treatment of cancers, such as breast cancer and mantle cell lymphoma, as well as inflammatory diseases like colitis. Various delivery systems, including nanoliposomes and targeted stabilized nanoparticles (tsNPs) conjugated with antibodies (e.g., anti-β7 integrin), have been explored to improve its stability and cell-specific uptake.
