Overview

Cyclin E1 MGD is a molecular glue degrader (MGD) being developed by TRIANA Biomedicines. It is designed to target Cyclin E1 (CCNE1), a key regulator of the G1/S phase transition in the cell cycle that is frequently amplified or overexpressed in various malignancies. As a molecular glue, the compound induces a novel interaction between Cyclin E1 and an E3 ubiquitin ligase, leading to the selective ubiquitination and subsequent proteasomal degradation of the Cyclin E1 protein. This therapeutic strategy is intended to address CCNE1-amplified solid tumors and hormone receptor-positive (HR+), HER2-negative breast cancer, particularly in cases where Cyclin E1 overexpression contributes to resistance against standard-of-care treatments like CDK4/6 inhibitors. The program is currently in the lead optimization stage of preclinical development.
