Cyclo-(histidyl-proline) (CHP) is an endogenous, blood-brain barrier-permeable cyclic dipeptide derived from the proteolytic cleavage of thyrotropin-releasing hormone (TRH). It acts as a neuroprotective and anti-inflammatory agent by modulating key cellular signaling pathways. Specifically, CHP activates the Nuclear factor erythroid 2-related factor 2 (Nrf2) pathway, which enhances the antioxidant response, and inhibits the nuclear accumulation of Nuclear factor-kappa B (NF-κB), thereby reducing pro-inflammatory cytokine production. While it has demonstrated potential in preclinical models of neurodegeneration, metabolic disorders, and inflammation, it is currently utilized primarily as a research reagent and has no established clinical development program or commercial sponsor.
