This is a combination chemotherapy regimen consisting of three drugs: - Cyclophosphamide, an alkylating agent that interferes with DNA replication and RNA transcription in cancer cells. - Doxorubicin (also known as Adriamycin), an anthracycline antibiotic that intercalates DNA and inhibits topoisomerase II, leading to inhibition of DNA replication and generation of free radicals. - Dacarbazine (also known as imidazole carboxamide or DTIC), a triazene derivative that acts as an alkylating agent after metabolic activation, causing methylation of guanine bases in DNA. This combination has been studied for the treatment of advanced malignant mesothelioma[1] and metastatic soft tissue sarcoma[4]. The addition of dacarbazine to the standard cyclophosphamide-doxorubicin regimen did not show significant improvement in response rates or survival compared to regimens without it. Toxicities are significant and include myelosuppression among others. Each component is approved for use in various cancers; however, this specific three-drug combination is not widely used today due to limited benefit over two-drug regimens.
