This is an investigational multi-agent combination regimen consisting of five drugs: - **Cyclophosphamide** (an alkylating agent that cross-links DNA to prevent cell division) - **Doxorubicin** (an anthracycline antibiotic that intercalates DNA and inhibits topoisomerase II) - **Durvalumab** (a monoclonal antibody immune checkpoint inhibitor targeting PD-L1) - **Oleclumab** (a monoclonal antibody targeting CD73 to modulate the tumor microenvironment and enhance anti-tumor immunity) - **Paclitaxel** (a microtubule-stabilizing agent that disrupts mitosis) This combination brings together cytotoxic chemotherapy agents with two immunotherapies. Cyclophosphamide, doxorubicin, and paclitaxel are established chemotherapeutics for breast cancer and other solid tumors[2][6][7]. Durvalumab is approved for several cancers as a PD-L1 inhibitor; oleclumab is an experimental anti-CD73 antibody designed to further augment immune response against tumors. The rationale behind this regimen is to combine direct cytotoxic effects on tumor cells with enhanced anti-tumor immunity.
