The combination of cyclophosphamide, doxorubicin, and nab-paclitaxel is a neoadjuvant chemotherapy regimen primarily investigated for the treatment of high-risk breast cancer subtypes, including locally advanced breast cancer (LABC), inflammatory breast cancer (IBC), and triple-negative breast cancer (TNBC). This regimen typically utilizes a sequential or concurrent administration strategy. Nab-paclitaxel, a nanoparticle albumin-bound formulation of paclitaxel, is employed as a solvent-free alternative to conventional taxanes to enhance intratumoral drug delivery via albumin-mediated transport mechanisms and to reduce solvent-related toxicities. The regimen combines the DNA-alkylating properties of cyclophosphamide, the topoisomerase II inhibition and DNA intercalation of doxorubicin, and the microtubule-stabilizing effects of nab-paclitaxel to induce pathologic complete response (pCR) in aggressive breast malignancies.
