This is a combination therapy consisting of cyclophosphamide, doxorubicin, and vitamin D. Cyclophosphamide is an alkylating agent that interferes with DNA replication in cancer cells, leading to cell death. Doxorubicin is an anthracycline antibiotic that intercalates into DNA, inhibits topoisomerase II activity, generates free radicals causing DNA damage, and induces apoptosis in rapidly dividing cells. Vitamin D (including its analogs such as cholecalciferol or calcitriol) acts primarily through the vitamin D receptor to regulate gene expression; it has been investigated for its potential anti-inflammatory effects and ability to reduce chemotherapy-induced toxicity (notably cardiotoxicity from doxorubicin), as well as possible synergistic antitumor effects when combined with cytostatics[1][2][6]. This combination has been studied mainly in breast cancer settings where standard adjuvant regimens include cyclophosphamide plus doxorubicin ("AC"), with added vitamin D supplementation for potential cardioprotection or enhanced efficacy[1][6]. The combination may also be explored preclinically for other cancers.
