Overview

This is a combination chemotherapy regimen consisting of three agents: - **Cyclophosphamide** is an alkylating agent that interferes with DNA replication and transcription by cross-linking DNA strands, leading to cell death. - **Epirubicin** is an anthracycline antibiotic that intercalates into DNA and inhibits topoisomerase II, preventing DNA repair and synthesis. It also generates free radicals that damage cellular components. - **Trastuzumab** is a monoclonal antibody targeting the human epidermal growth factor receptor 2 (HER2). By binding to HER2 on cancer cells, it blocks downstream signaling pathways involved in cell proliferation and survival. This combination has been studied primarily for the treatment of HER2-positive metastatic breast cancer (MBC) as well as in neoadjuvant/adjuvant settings. The rationale for using epirubicin instead of doxorubicin is its lower cardiotoxicity profile when combined with trastuzumab[1][2]. The regimen has demonstrated promising efficacy with manageable cardiac safety compared to regimens containing doxorubicin[1][2].
