Overview

Cyclophosphamide + eribulin is an investigational combination chemotherapy regimen studied primarily for the treatment of advanced or metastatic breast cancer. Cyclophosphamide is an alkylating agent that crosslinks DNA, leading to cell death, while eribulin is a microtubule dynamics inhibitor that binds to the plus ends of microtubules, preventing their elongation and causing irreversible mitotic arrest. Eribulin also has additional effects on the tumor microenvironment, including reversal of epithelial-to-mesenchymal transition (EMT) and remodeling tumor vasculature to improve perfusion. The combination has shown encouraging clinical benefit rates in heavily pretreated patients with advanced breast cancer in phase Ib/II studies, though it remains experimental and not FDA-approved as a fixed combination[1][2][3][5].
