This is a combination chemotherapy regimen consisting of three cytotoxic agents—cyclophosphamide, etoposide, and mitoxantrone. Cyclophosphamide is an alkylating agent that interferes with DNA replication and RNA transcription by cross-linking DNA strands. Etoposide is a topoisomerase II inhibitor that induces DNA strand breaks by stabilizing the temporary break made by the enzyme during DNA replication. Mitoxantrone is an anthracenedione antineoplastic agent that intercalates into DNA and inhibits both topoisomerase II activity and DNA synthesis. This combination has been investigated primarily for use in hematologic malignancies such as refractory lymphoma (including as part of high-dose regimens prior to autologous bone marrow transplantation) and has also been used in other settings where aggressive cytoreduction is required[1][9]. The regimen aims to maximize tumor cell kill through synergistic mechanisms while balancing toxicity.
