Overview

Cyclophosphamide + fludarabine is a combination chemotherapy regimen primarily used in the treatment of chronic lymphocytic leukemia (CLL) and other hematologic malignancies. Cyclophosphamide is an alkylating agent that cross-links DNA strands, inhibiting DNA replication and leading to cell death. Fludarabine is a purine analog antimetabolite that inhibits DNA polymerase alpha, ribonucleotide reductase, and DNA primase—disrupting DNA synthesis and inducing apoptosis in both proliferating and non-proliferating cells. The combination exploits synergistic cytotoxicity to improve response rates compared to either drug alone[2][3][5][8]. This regimen may be referred to as "FC" or "FLU-CY" in clinical practice.
