Drug intelligence / Profile preview
The combination of cyclophosphamide, fludarabine, and albumin-bound paclitaxel (commonly referred to as the FNC regimen) is an innovative preconditioning and lymphodepletion regimen developed primarily by CARsgen Therapeutics. It is administered prior to the infusion of chimeric antigen receptor (CAR) T-cell therapies (such as satricabtagene autoleucel) for solid tumors, including advanced gastric, gastroesophageal junction, and pancreatic cancers. While the conventional cyclophosphamide and fludarabine (Cy/Flu) regimen is the standard for lymphodepletion in hematological malignancies, the addition of low-dose albumin-bound paclitaxel (nab-paclitaxel) is designed to address the unique challenges of solid tumors. Nab-paclitaxel helps deplete the dense desmoplastic stroma, reduces immunosuppressive myeloid-derived suppressor cells (MDSCs), and enhances the infiltration, expansion, and persistence of CAR T-cells within the immunosuppressive tumor microenvironment.
