This is a multi-agent combination regimen consisting of five drugs: cyclophosphamide, fludarabine, cyclosporin, alemtuzumab (also known as CAMPATH-1H), and granulocyte-macrophage colony-stimulating factor (GM-CSF). Cyclophosphamide is an alkylating agent that interferes with DNA replication. Fludarabine is a purine analog that inhibits DNA synthesis. Cyclosporin is an immunosuppressant that inhibits calcineurin and T-cell activation. Alemtuzumab is a monoclonal antibody targeting CD52 on lymphocytes, leading to cell lysis. GM-CSF stimulates the production of white blood cells and enhances immune function. This combination has been studied in the context of hematopoietic stem cell transplantation conditioning regimens and for certain hematologic malignancies[4][8].
