Cyclophosphamide + interferon-gamma is an investigational combination therapy consisting of the alkylating agent cyclophosphamide and the cytokine immunomodulator interferon-gamma. Cyclophosphamide is a nitrogen mustard antineoplastic and immunosuppressive agent that acts primarily by cross-linking DNA, leading to cell death in rapidly dividing cells. Interferon-gamma is a recombinant cytokine that activates macrophages, enhances antigen presentation, and modulates immune responses. The combination has been explored in oncology settings—particularly as part of lymphodepletion regimens prior to adoptive cell transfer (ACT) or as an adjunct to chemotherapy for advanced cancers such as ovarian cancer—with the aim of enhancing anti-tumor immunity by both direct cytotoxic effects (cyclophosphamide) and immune activation (interferon-gamma). Clinical studies have reported both potential efficacy benefits and significant safety concerns when these agents are combined[3][4].
